Zombie Slayer is open from the start and is the easiest. A Tier II Revenant Horror kill unlocks Spider, Tier II Tarantula Broodfather unlocks Wolf, Tier IV Sven Packmaster unlocks Enderman, and Tier III Voidgloom Seraph unlocks Blaze. Vampire Slayer is in the Rift and can be started almost any time.
